Yeast Expression

Boehringer Ingelheim BioXcellence™ has developed and implemented a spectrum of processes employing yeast hosts. With our expertise we are able to generate suitable fermentation strains and downstream processes in terms of economy, product quality and efficiency.

 

  • At Boehringer Ingelheim Hansenula polymorpha fermentations were developed that achieved titers above 5 g/L.
  • Saccharomyces cerevisiae processes with protein titers up to 3 g/L were designed and implemented in our large-scale production plant at Vienna.

 

Furthermore, Boehringer Ingelheim BioXcellence™ has access to the proprietary Pichia pastoris expression system via collaboration with VTU Technology www.vtu-technology.com.

The technology platform is characterized by an unparalleled speed of development, high yields and high-quality results thanks to the application of advanced and exclusive technologies.

The proteins are secreted into the fermentation supernatant. A strong and inducible AOX promoter and variants of it maximize expression of more than 10 g/l. This technology is best suited for the expression of e.g. Nanobodies® and HSA fusion proteins.

The key features of the Pichia pastoris expression system are as follows:

  • High-speed expression strain development by VTU and scale-up, and GMP manufacturing by Boehringer Ingelheim BioXcellence
  • Expression titers of more than 10 g/L
  • Purified protein in as little as 12 weeks
